Bu çalışmada, mercimek (Lens culinaris Medik.) bitkisinin genomuna ait bağlantı (linkaj) haritasına SSR (Simple Sequence Repeats) markörlerinin katılımı amaçlanmıştır. Genom haritası oluşturulmasında WA8649041 ve Precoz ebeveynlerin çaprazlanmasıyla elde edilen 93 adet rekombinant saf hat (Rekombinant Inbred Lines, RILs) populasyonu kullanılmıştır. Çalışmada 175 SSR primeri kullanılmış, bu primerlerden 25 adedi polimorfik, 109 adedi monomorfik, 41 adedi çalışmamıştır. Polimorfik olduğu tespit edilen SSR markörlerinden, 13 adedi haritalanmış, haritalanan markörler, genetik bağlantı haritasında belirli bir kromozom veya grupta toplanmayıp farklı bağlantı gruplarına dağılmıştır. Hali hazırda mevcut 123 markörden oluşan genom haritasına (57 AFLP, 48 RAPD, 16 ISSR ve 2 morfolojik markör), bu çalışmayla 13 SSR markör ilave edilerek genom haritasındaki toplam markör sayısı 136 olmuştur. Genom haritası 11 bağlantı (linkaj) grubundan oluşmuş, toplam harita uzunluğu 1311.2 cM ve iki markör arası ortalama uzunluk 9.64 cM olarak belirlenmiştir.
The objective of this study was to integrate the SSR (Simple sequence Repeats) markers in lentil (Lens culinaris Medik.) genetic linkage map. To construct the linkage map, an F6 derived Recombinant inbreed line (RIL) population with 93 lines developed from the cross of WA8649041 x Precoz was used. Linkage map was comprised of RAPD, ISSR, AFLP, ISSR and morphological markers. One hundred seventy five (175) SSR markers were used in the study. 25 SSR markers were polymorphic, while 109 SSR markers were monomorphic and 41 SSR markers did not work. SSR markers were distributed in different linkage groups rather than clustering in some linkage groups. Of the 21 SSR markers, 13 of them were mapped in lentil genome and the current genetic linkage map is comprised of 136 markers including 57 AFLP, 48 RAPD, 16 ISSR, 2 morphological and 13 SSR markers. Genetic linkage map is comprised of 11 linkage groups with 1311.2 cM in length and average genetic distance between two markers is 9.64 cM.
